Credit: Pinngortitaleriffik – Greenland Nature Institute Local hunters in the Icefjord near Ilulissat are familiar with a special variety of ringed seal known as the Kangia seal. Distinct in size and appearance, the Kangia seal is considerably larger and boasts a notably different fur color and pattern compared to the common Arctic ringed seals. Recent scientific research has revealed that this distinctive seal has been genetically isolated from its Arctic counterparts for a long period of time – more than 100,000 years. Exploring the Arctic’s natural wonders can be difficult. The extreme weather and vast distances often hinder researchers in their quest to uncover nature’s mysteries. However, a research project, led by Greenlandic and Danish researchers, has now succeeded in describing a new type of ringed seal that lives in the Icefjord near Ilulissat in West Greenland; a unique natural area on the UNESCO World Heritage List. The results have recently been published in the renowned scientific journal Molecular Ecology. The Kangia ringed seal is larger than the typical Arctic ringed seal and its fur color is different with more distinctive patterns. Credit: Pinngortitaleriffik – Greenland Nature Institute A small population Over a number of years, the researchers together with local hunters captured seals in nets and mounted a small satellite transmitter on the seals’ backs. When the seals were up for air, the satellite transmitter sent a message about their location. ”We could see that the Kangia seals primarily stay inside the Icefjord. We were able to count the seals from a plane and therefore able to estimate that there are only approx. 3,000 of these special Kangia ringed seals,” says Aqqalu Rosing-Asvid, Senior Researcher at the Pinngortitaleriffik – Greenland Institute of Nature, and one of the researchers behind the study. Credit: Pinngortitaleriffik – Greenland Nature Institute The small resident population is highly unusual compared to the typical Arctic ringed seal, which has an enormous population size and often travels thousands of kilometers around the Arctic in search of food. Isolated for thousands of years The researchers also took small tissue samples from the captured seals. The samples were sent for genetic analyses to uncover the seals’ DNA profile, and the results revealed that the Kangia ringed seals are genetically different from the typical Arctic ringed seals. Perhaps also special seals in other Arctic fjords The study emphasizes that there is still much we do not know about the diversity of organisms in the Arctic and thus their possibilities to adapt to climate change and human activities. “There are many other fjords in the Arctic that have not yet been studied in detail, and where the ringed seals may also have locally developed new genetic variants,” points out Rune Dietz, Professor at the Department of Ecoscience at Aarhus University, who also participated in the study. Credit: Maggie Johnson © Woods Hole Oceanographic Institution In September of 2017, Woods Hole Oceanographic Institution postdoctoral scholar Maggie Johnson was conducting an experiment with a colleague in Bocas del Toro off the Caribbean coast of Panama. After sitting on a quiet, warm open ocean, they snorkeled down to find a peculiar layer of murky, foul-smelling water about 10 feet below the surface, with brittle stars and sea urchins, which are usually in hiding, perching on the tops of coral. This unique observation prompted a collaborative study explained in a new paper published on July 26, 2021, in Nature Communications analyzing what this foggy water layer is caused by, and the impact it has on life at the bottom of the seafloor. All of the macro-organisms are trying to get away from this deoxygenated water, and those that cannot escape essentially suffocate. I have never seen anything like that on a coral reef,” said Johnson. The study looks closely at the changes occurring in both coral reef and microbial communities near Bocas del Toro during sudden hypoxic events. When water drops below 2.8mg of oxygen per liter, it becomes hypoxic. More than 10% of coral reefs around the world are at high risk for hypoxia (Altieri et al. 2017- tropical dead zones and mass mortalities on coral reefs). Credit: Maggie Johnson © Woods Hole Oceanographic Institution “There is a combination of stagnant water from low wind activity, warm water temperatures, and nutrient pollution from nearby plantations, which contributes to a stratification of the water column. From this, we see these hypoxic conditions form that start to expand and infringe on nearby shallow habitats,” explained Johnson. Investigators suggest that loss of oxygen in the global ocean is accelerating due to climate change and excess nutrients, but how sudden deoxygenation events affect tropical marine ecosystems is poorly understood. Past research shows that rising temperatures can lead to physical alterations in coral, such as bleaching, which occurs when corals are stressed and expel algae that live within their tissues. If conditions don’t improve, the bleached corals then die. However, the real-time changes caused by decreasing oxygen levels in the tropics have seldom been observed. Credit: Maggie Johnson © Woods Hole Oceanographic Institution At a local scale, hypoxic events may pose a more severe threat to coral reefs than the warming events that cause mass bleaching. These sudden events impact all oxygen-requiring marine life and can kill reef ecosystems quickly. Investigators reported coral bleaching and mass mortality due to this occurrence, causing a 50% loss of live coral, which did not show signs of recovery until a year after the event, and a drastic shift in the seafloor community. The shallowest measurement with hypoxic waters was about 9 feet deep and about 30 feet from the Bocas del Toro shore. What about the 50% of coral that survived? Johnson and her fellow investigators found that the coral community they observed in Bocas del Toro is dynamic, and some corals have the potential to withstand these conditions. This discovery sets the stage for future research to identify which coral genotypes or species have adapted to rapidly changing environments and the characteristics that help them thrive. Credit: Maggie Johnson © Woods Hole Oceanographic Institution Investigators also observed that the microorganisms living in the reefs restored to a normal state within a month, as opposed to the macro-organisms, like the brittle stars, who perished in these conditions. By collecting sea water samples and analyzing microbial DNA, they were able to conclude that these microbes did not necessarily adjust to their environment, but rather were “waiting” for their time to shine in these low-oxygen conditions. Credit: Maggie Johnson © Woods Hole Oceanographic Institution “The take home message here is that you have a community of microbes; it has a particular composition and plugs along, then suddenly, all of the oxygen is removed, and you get a replacement of community members. They flourish for a while, and eventually hypoxia goes away, oxygen comes back, and that community rapidly shifts back to what it was before due to the change in resources. This is very much in contrast to what you see with macro-organisms,” said Jarrod Scott, paper co-author and postdoctoral fellow at the Smithsonian Tropical Research Institute in the Republic of Panama. Scott and Johnson agree that human activity can contribute to the nutrient pollution and warming waters which then lead to hypoxic ocean conditions. Activities such as coastal land development and farming can be better managed and improved, which will reduce the likelihood of deoxygenation events occurring. The study provides insight to the fate of microbe communities on a coral reef during an acute deoxygenation event. Reef microbes respond rapidly to changes in physicochemical conditions, providing reliable indications of both physical and biological processes in nature. The shift the team detected from the hypoxic microbial community to a normal condition community after the event subsided suggests that the recovery route of reef microbes is independent and decoupled from the benthic macro-organisms. This may facilitate the restart of key microbial processes that influence the recovery of other aspects of the reef community. A devastating fungus affecting amphibians worldwide may be countered by a newly discovered virus that infects it. Researchers at UC Riverside are exploring how this virus could be engineered to slow down or stop the spread of the fungal disease, potentially saving numerous amphibian species from decline or extinction. Scientists have discovered a virus that infects a deadly amphibian fungus, offering hope for combating a disease that has led to significant declines in frog and toad populations globally. A fungus devastating frogs and toads on nearly every continent may have an Achilles heel. Scientists have discovered a virus that infects the fungus, and that could be engineered to save the amphibians. The fungus, Batrachochytrium dendrobatidis or Bd, ravages the skin of frogs and toads, and eventually causes heart failure. To date it has contributed to the decline of over 500 amphibian species, and 90 possible extinctions including yellow-legged mountain frogs in the Sierras and the Panamanian golden frog. Virus as a Potential Savior A new paper in the journal Current Biology documents the discovery of a virus that infects Bd, and which could be engineered to control the fungal disease. The UC Riverside researchers who found the virus are excited about the implications of their discovery. In addition to helping them learn about how fungal pathogens rise and spread, it offers the hope of ending what they call a global amphibian pandemic. If their populations all over the world collapse, it could be devastating,” said UCR microbiology doctoral student and paper author Mark Yacoub. “They’re also the canary in the coal mine of climate change. As temperatures get warmer, UV light gets stronger, and water quality gets worse, frogs respond to that. If they get wiped out, we lose an important environmental signal,” Yacoub said. Genetic Research and Discoveries Bd was not prevalent before the late 1990s, but then, “all of a sudden frogs started dying,” Yacoub said. When they found the Bd-infecting virus, Yacoub and UCR microbiology professor Jason Stajich had been working on the population genetics of Bd, hoping to gain a better understanding about where it came from and how it is mutating. Credit: Mark Yacoub/UCR Deciphering Viral and Fungal Interactions “We wanted to see how different strains of fungus differ in places like Africa, Brazil, and the U.S., just like people study different strains of COVID-19,” Stajich said. To do this, the researchers used DNA sequencing technology. As they examined the data, they noticed some sequences that did not match the DNA of the fungus. “We realized these extra sequences, when put together, had the hallmarks of a viral genome,” Stajich said. Previously, researchers have looked for Bd viruses but did not find them. The fungus itself is hard to study because complex procedures are required to keep it alive in a laboratory. The Challenge of Studying Bd and Its Virus “It is also a hard fungus to keep track of because they have a life stage where they’re motile, they have a flagellus, which resembles a sperm tail, and they swim around,” Stajich said. Additionally, the virus that infects Bd was hard to find because most known viruses that infect fungi, called mycoviruses, are RNA viruses. However, this virus is a single-stranded DNA virus. Credit: Mark Yacoub/UCR Future Research and Amphibian Immunity It appears that only some strains of the fungus have the virus in their genome. But the infected ones seem to behave differently than the ones that don’t. “When these strains possess the virus they produce fewer spores, so it spreads more slowly. But they also might become more virulent, killing frogs faster,” Stajich said. Right now, the virus is essentially trapped inside the fungal genome. The researchers would eventually like to clone the virus and see if a manually infected strain of Bd also produces fewer spores. “Because some strains of the fungus are infected and some are not, this underscores the importance of studying multiple strains of a fungal species,” Yacoub said. Moving forward, the researchers are looking for insights into the ways that the virus operates. “We don’t know how the virus infects the fungus, how it gets into the cells,” Yacoub said. “If we’re going to engineer the virus to help amphibians, we need answers to questions like these.” In some places, it appears there are a few amphibian species acquiring resistance to Bd. “Like with COVID, there is a slow buildup of immunity.
